The Institute of Genetic Medicine at Johns Hopkins University School of Medicine has a post-doctoral researcher opening to conduct research on the Identification and Characterization of Small Molecules for Lysosomal Storage Diseases.

The primary responsibility is the development of a high-throughput screening (HTS) assay to identify small molecules that are able to assist the folding of misfolded mutant lysosomal enzymes that are deficient in different lysosomal storage diseases (e.g. metachromatic leukodystrophy and Krabbe disease). The postdoc will work on developing a robust and innovative cell-based HTS assay. Once the assay is implemented, the postdoc is expected to screen small molecule libraries including FDA-approved and drug-like compounds. In addition, the postdoc will develop counterscreening and secondary assays. Lastly, the postdoc will characterize and validate the candidate compounds from the screening using cell lines from different patients and eventually animal models. The post-doctoral researcher should have completed a PhD and/or MD degrees (PhD in the field of biomedical sciences) and have experience in the following areas: enzymatic and protein time-sensitive assays; basic molecular biology techniques including but not limited to molecular cloning, transfection, infection, RNA isolation, and Northern blotting; perform tissue culture procedures; knowledge of specific softwares such as GraphPad, Excel, PubChem, PDB, Volocity and ZEN 2009 Edition; basic proteomics techniques including enzyme kinetics, subcellular fractionation, immunoprecipitation, immunoblots, pulse-chase assays, confocal and live cell microscopy, immunofluorescence assays, etc.

The post-doctoral researcher will interact with members of HTS centers from the Molecular Libraries Probe Production Centers, a network of national laboratories, including NIH Chemical Genomics Center, Rockville, MD. In addition, the postodoc will have the opportunity to work and interact with different leading researchers and trainees from the IGM and present their work at national and international conferences.
